Intelligent Biomedical Optical Imaging and Diagnostics
By converging nanophotonics and biomedical imaging, our research interests focus on developing novel photonic devices with enhanced optical sensing and imaging capabilities and emerging imaging technologies to break current limitation in optical imaging and tackle evolving biomedical challenges.
Super-Resolution Light Microscopy emerges as a powerful technology that offers the imaging resolution far exceeding the diffraction limit of light, enabling researchers to “see” the extremely fine details previously deemed unresolvable by conventional optical microscopy. Despite the great success, current advancements of SRLM have yet to harness high-throughput analytical capabilities to reveal the unique physical and chemical properties of individual biomolecules from their assembly. Aiming to overcome the diffraction barrier of analytical imaging methods, it is our interest to develop enhanced multifunctional super-resolution microscopy availing multimodal single-moleucle spectroscopy.

In order to efficiently acquire molecular specific contrasts by optical means, improvements in detection sensitive and measurement precision are essential for sensing subtle signals in diverse forms. Emerged as a promising technology, micro-/nano-photonics offers unlimited potentials in designing and engineering novel optical devices for sensing, imaging, and signal processing by utilizing its extraordinary capability in controlling electromagnetic waves with sub-wavelength scale photonic structures.

Multiphoton microscopy has become an essential tool in brain science research, offering the ability to observe deep structural connections and functional activities within the brains of living animals. Recent advancements have highlighted the critical need for advanced neuronal imaging and modulation technologies that provide high spatial and temporal resolution at mesoscopic scales. We develop cutting-edge multiphoton imaging and holographic optogenetic stimulation system, integrating with deep learning-based imaging and data processing methods. This system not only captures large-field, high-resolution images of deep tissues in vivo but also enables real-time analysis and precise regulation of neuronal activities. Its capability to synchronize and regulate these activities in real time positions it as an ideal platform for conducting closed-loop neuroscience research at mesoscopic scales, paving the way for novel insights and methodologies in Neuroscience.

Since physiological functions account for amazingly rich spatial/time scales and processes, multi-scale and multi-physics approaches are essential for investigating the complex, nonlinear relationship with the emerging shapes and functions of cells, tissues, and organs in living organisms. While super-resolution light microscopy is powerful in understanding the detailed biological mechanisms that underlie disease at molecular scale, system scale optical imaging technologies can then provide visualization of pathophysiology at spatial scales from the subcellular to whole organ levels. On the other hand, integrating multiply imaging and spectroscopic modalities has been demonstrated highly beneficial in creating comprehensive research and diagnostic platforms. For example, optical coherence tomography, photoacoustic microscopy, and confocal microscopy can offer high-resolution volumetric morphological details, whereas near-infrared spectroscopy, Raman spectroscopy, and fluorescence microscopy are highly sensitive to chemical signatures.
